Complete genome analysis of the newly isolated Shigella sonnei phage vB_SsoM_Z31

This work describes the characterization and genome annotation of newly isolated lytic phage vB_SsoM_Z31 (referred to as Z31), from wastewater samples collected in Dalian, China. Transmission electron microscopy revealed that Z31 belongs family Myoviridae, order Caudovirales. specifically infects Shigella sonnei, dysenteriae, Escherichia coli. The is an 89,355-bp-long dsDNA molecule with a G+C content 38.87%. It was predicted contain 133 ORFs encode 24 tRNAs. No homologs virulence factor genes or antimicrobial resistance were found this phage. Based on results nucleotide sequence alignment phylogenetic analysis, assigned genus Felixounavirus, subfamily Ounavirinae.

The properties of three T-related phages-35, 55, and 3201-isolated from Shigella sonnei were studied. They were similar with respect to morphology of plaques, duration of the latent periods, lysis inhibition effect, and serological characteristics. These phages closely resembled the T-even phages. Phages 3201, 35, and 55 had the same host range and receptor specificity as phage T2.

The isolate contained a plasmid-borne mph(A) gene encoding a macrolide 2 -phosphotransferase that inactivates macrolides. Emergence of the mph(A) gene in S. sonnei may limit usefulness of azithromycin for treatment of shigellosis.
